Empower Tip: Review Window and the Processing Method

Question: how do I subtract a baseline at a particular wavelength from my PDA data? We need to first save the blank as 2D data at the wavelength of interest and the rest is easy.

We begin by bringing a sample set into Review, extract the wavelength of interest and observe the baseline of one of the injections. We see the rise in the baseline due to the gradient.

First, we need to give the blank injection a label either in the Sample Set method prior to acquiring the data or post-run in Alter Sample.

Select the blank injection and from the File menu select Save Chromatogram.

Similar to what we did in Tip 49, Derive a Channel and on the First (Only) Channel tab select PDA: Single Wavelength from the Channel drop-down list.

Enter the wavelength, in this example it is 215.

On the Second Channel tab select subtraction from the Operator drop down list and then type 215.0nm (this is how Empower names the 2D data as per step 3) into the Channel field, do not select anything from the Channel drop down list.

Check the From injection labeled box and enter the label for the blank injection.

Click OK, give the new Derived Channel a name and select the Processing Method in the Channel table within the Method Set.

Apply the Method Set to the same injection as in step 1 and zoom in on the corrected baseline.

It’s that easy!
