Tip #58: Deriving a Channel in Empower (part 11)

In this tip, we will continue the discussion on Deriving a TIC plot from 3D MS data, specifically how to improve the noisy TIC plot.

You may recall the previous tip (Tip 57) when we derived the TIC Plot, it was noisy and I mentioned that it could be improved. The Component Detection Algorithm, or CODA, is a method that was specifically developed for LC/MS data in order to reduce random noise and high background so as to simplify interpretation of the data. CODA retains mass chromatograms which contain information (i.e., analyte peaks) in the data set and removes mass chromatograms which add noise to the TIC plot.

We begin by Deriving the TIC plot as we learned in Tip 57.

Return to the Method Set window, right click on the Derived Channel in the tree and select Properties.

Click the MS CODA tab, enable CODA and click OK.

STEP 4

Return to the Main Window, re-apply the Method Set and notice the improvement in the TIC plot.
