In this tip for Empower Chromatography Data System (CDS), we resume our series on Calculating Amount/Concentration and review Calibration Curve fit types.
Let’s start with some background information.
There are a variety of different fit types for Calibration Curves which can be found in the Processing Method on the Components tab. They are as follows:
Single Level Calibration: linear thru zero and response factor
Multilevel Calibration Matrix Operations:
Multilevel Calibration: linear, inverse linear, log-log linear, quadratic, cubic, fourth order, fifth order and power curve
Multilevel Forced Thru Zero: linear, quadratic, cubic, fourth order, fifth order and response factor
Multilevel Calibration: point-to-point and cubic spline
Linear thru zero is defined by the equation ‘y = Bx’.
STEP
The amount or concentration for the component in the sample is calculated by the equation ‘x =y/B’.

Response factor is defined by the equation ‘RF = Y Value/X Value’.
The RF is the slope of the Calibration Curve. If multiple points are used, the RF for each individual point is determined and the average RF is used as the slope of the curve.
