In this tip, we will discuss creating an Instrument Method for SEC-MALS
Let’s get started.
When we create a new Instrument Method, these are the default parameters for the DAWN™ on the General tab.
The Collection Interval has been adjusted in the final method. Some detectors have a Sampling Rate in points/second. This detector has a Collection Interval. Set the value so it results in a sufficient number of points across the peak for reproducible retention time, area and height calculations. I generally recommend 15-20 points across an HPLC peak and 20-25 points across a UPLC peak.
These are the default parameters on the Misc tab which are fine for our purposes.
These are the default parameters on the Data tab which are fine for our purposes. It is important to enable the Laser Monitor because the light scattering signals are divided by the laser monitor which corrects for fluctuations in the laser intensity due to laser power fluctuations.
